A Novel Approach for Calculating Exact Forms of mRNA Distribution in Single-Cell Measurements

Gene transcription is a stochastic process manifested by fluctuations in mRNA copy numbers individual isogenic cells. Together with mathematical models of transcription, the massive distribution data that can be used to quantify levels fitted Pm(t), which probability producing m molecules at time t single cell. Tremendous efforts have been made derive analytical forms rely on solving infinite arrays master equations models. However, current approaches focus steady-state (t→∞) or require several parameters zero infinity. Here, we present an approach for calculating Pm(t) time, where all are positive and finite. Our was successfully implemented classical two-state model widely three-state may further developed different constant kinetic rates transcription. Furthermore, direct computations showed regulations gene activation generate discriminated dynamical bimodal features under same similar distribution.
